The Fret Mill on Roanoke's city market has a pretty good selection of guitars and usually a few mandolins of intrest, some other stores in the nearby would be Franklin music on 220 south about 15 miles out and Halls music in Rocky Mount another 10 miles or so south on 220.
I'll second all mandoplums choices adding Kelly's Music, also in Roanoke. There's a decent shop off the beaten path in Greensboro but I've lost the name. If you have time dive into Lexington, Va and have lunch plus visit Rockbridge Music while you are in that fine little town (John Schofield 's wife owns that store. He has a history of creating great mandos).
Fretwell Music in Staunton, Va. carries a variety of instruments, & they are known for their basses..I have picked some very nice guitars and mando's there too. Check out their website.

If you are going to Roanoke on the way back, and it's a friday, check out downtown Floyd, Va for the Friday night Jamboree. Depends on weather, but if its warm enough the street outside the Generals Store (Rte 8 from 81) gets filled with multiple groups of musicians jamming away while there is paid entertainment going on inside the store.

Second the Floyd jam. Great time. Wayne Henderson's shop is near there, too. He's a guitar builder mainly but also builds killer mandolins. Also a great picker.
Another spot further south is the Carter fold outside of Bristol.

Check for Prissy Polly's in Winston for some great Q and some great grass, depending on their schedule.
Capo Music in Abingdon has acoustic instruments. It's close to i81.

Prissy Polly's is a great tip as Alan suggests, really good barbecue and you can catch some live Bluegrass music. But it's in Kernersville NC, about 15 miles east of Winston-Salem, just off I-40 Business
Jackson's Music in Winston Salem is a nice little shop.
You might contact Bryce (David Houchens) and see if he has something for you to test drive. He's just south of Charlottesville. If you go through ROANOKE on a wed. night ( 6PM- 9PM) you could check out some friends of mine at The Coffee Pot. Traditional bluegrass/ gospel done by some pretty good pickers. Very cool historic music venue too. Have fun!
