I have installed frets on the fingerboard before gluing it down to the neck. There is a bit of a twist, maybe 3/16". When I glue it down should there be any concern that the frets can loosen as it is clamped back flat?

You might try dampening the fretboard and clamping it to the workbench for a day or so. The StewMac video even shows putting a light over it, to apply gentle heat as it dries. It should flatten out. Then you can see if the frets are still OK.

Putting the frets in the slots tends to force the slots wider and this results in a bow in the fingerboard.
Clamp it to the benchtop for a couple of days. This will force the frets to seat in the slots better and take out much of the bend.
If all the bow does not come out, I'd still glue it up to the neck. Then check the frets and level as needed.
